You can use the Settings in FF Metro mode to update Nightly.
But the update process does not complete from Metro.
You must go into Desktop mode and manually (there are prompts) finish the update.

STRs - fairly obvious
Make sure FF is the default browser program
Open FF in Metro mode
Swipe from the right and select Settings
Select About
Select Check for Updates
Get the update

FF will start the update process and close in metro mode
Note that it does not come back up at that point to finish the upgrade and launch

Move over to desktop mode
Finish the update from there
Summary: It does not appear to be possible to update FF from Metro mode → defect - It does not appear to be possible to update FF from Metro mode
Can you download the update and click to restart the browser from metro? If so that is not fixed yet and it's bug 874323.
I meant to also say,, but when you click the button nothing happens?
You need to manually click to start Firefox Metro again for the update to apply.
And then an additional time after the update is done.
Those extra restarts are being worked out in the already posted bugs.
Just tested upgrades on m-c and it works. I think you thought it wasn't working because you need to manually restart the metro interface after clicking to restart twice. The first restart is being tracked in bug 874323, the second restart will be posted separately.

Thanks James.

The issue of having to restart metro is being tracked. The report here in Comment 0 is about not being able to complete an update in Metro though, but you can, you just need to manually restart it by clicking the Metro tile yourself.
