Steps to reproduce:
Have Firefox 41.0 and my long-living profile. Let it to auto-update to 42.0.
Actual results:
Firefox 42.0 is unable to start. Eats one CPU core and does not show any window, infinitely.
It works with empty profile.
If I install back 41.0, all is working again. But it tries to auto-update with the same result.
If I install 42.0 manually, it does not work.
Expected results:
I expect newer version to work or at least complain if something is wrong with the profile.
If FF42 works with a fresh profile, so there is something wrong with your current profile. Maybe an add-on issue.

No, it does not allow me to switch to safe mode. Safe mode selection window does not appear.
In correction to my initial description, Firefox does not hang forever now. It crashes in about 3 minutes. I have filed two reports:

You can start in safe mode by using command line: cmd.exe > firefox.exe -safe-mode
firefox -safe-mode gives the same result: hangs for 3 minutes, eats all memory then crashes. One difference: this time, it did not allow me to report the crash.
I can confirm that copying places.sqlite and permissions.sqlite from my profile to fresh one makes it unusable. So this is definitely duplicate of 1221974.
