You cannot delete a locally installed extension until Firefox is closed.

Download an extension, say uBlock Origin, https://addons.mozilla.org/firefox/downloads/file/3521827/ublock_origin-1.25.2-an+fx.xpi, saving locally, say in C:\TMP.

New FF Profile
Drag extension into FF window to install it
Install it

Keep FF open

Result:

uBlock is installed (expected)
uBlock is in Profile/extensions/ (as [e-mail], expected)

Now attempt to delete, C:\TMP\ublock_origin-1.25.2-an+fx.xpi.

Result:

File In Use.
The action can't be completed because the file is in open in another program.
Close the file and try again.

Now, close FF.

Again attempt to delete, C:\TMP\ublock_origin-1.25.2-an+fx.xpi.

Result:

ublock_origin-1.25.2-an+fx.xpi is deleted as wanted.
