Steps to reproduce:
Opened Thunderbird 78.0.1
Actual results:
Nothing. No email accounts checked (no activity in the status bar)
Expected results:
New mails downloaded on start-up. Manual download works ok.

Wfm win10, IMAP, 78.0.1 (64-bit) - checks messages automatically at every startup (status bar flashes respective messages)
Can you check if the following is checked for all of your accounts?

Account settings > YourEmail@account > Server Settings > Check for new messages at startup

Accounts affected: POP or IMAP?

Does this only fail at first startup or at every startup?

Make sure to focus 100% on status bar whilst TB is starting - it's just a flash of less than a second. Strangely nothing in Activity manager after that (which I'd consider a bug).

I've checked all that you suggested and it all looks ok. Tried my wife's profile and that was ok. Created another profile for myself and I'm gradually starting to add my email accounts and all looks ok so far.
I obviously looks like a profile problem and I'm a bit annoyed that I didn't try that first before reporting it here.

Bug 1655645 is somewhat similar.
Sorry, markdown and me didn't agree on the formatting of my comment 1..., let me rephrase.
Could you pls answer these questions just in case we'll see this again (pls use reply feature and reply inline):
- Is "Check for new messages at startup" checked for all affected accounts?
- What is the server type of affected accounts: POP or IMAP?
- Does this only fail at first startup or at every startup?
- Anything in Error console (Ctrl+Shift+J) after it fails?
- Note that given that activity manager doesn't show this (bug?), and status bar flash is easily overlooked (and leaves no trace), this is best tested with new messages on server.

No worries, profile problems are tricky and not easy to blame here or there.
