Try running with -safe-mode (Help | Troubleshoot mode). May have to use the controls in the dialog that comes up to clear up issues if they still show.
I tried --safe-mode start-up and its optional disables but it did not work.
Send is normally grayed when Thunderbird is off-line or when there is no valid address to send to. One of these tests is broken, I guess.
New e-mails retrieval, by the way, works well as usual.

There is no general problem, so something wrong in your profile. Did you try resetting toolbars etc from the safe-mode dialog?
Having removed two addons (cardbook, mail-redirect) and resetting the tool bar had resolved "graying" of the send button.
However, send now and send later don't actually work.
It's a big issue for me now :-(
Thanks, Enoch.
