Thunderbird will not start up when "mail/read mail" is selected.  It will start
up if I do it from a shortcut.

Reproducible: Always
Steps to Reproduce:
1. open Firefox
2. Select "mail - read mail"
3. Nothing happens

Actual Results:  
Nothing.  Screen remains on Firefox, no HD activity.

Expected Results:  
Opened the Thunderbird mail program.
WFM on WinXP. 

Are you sure you have Thunderbird set as your default mail application?
