As I look at the menu, create filter from message is grayed out and disabled
Even when I selected an email file.

Reproducible: Always
Steps to Reproduce:
1.I get email
2.I  try to creat a message filter
3. I chose create filter from message

Actual Results:  
create filter from message is grayed out and disabled.

Expected Results:  
Try to fix it.

This is the wrong Product. Please change it to MailNews/Filters and don't forget
to check    "Reassign bug to owner and QA contact of selected component".

Does it mean a mail in a mail folder is selected or displayed?
Or an ".EML" file is opened from File/Open menu?

This problem has actually already been fixed.  The patch was checked in Oct 12; 
please try a more up-to-date nightly build.
