Since Thunderbird automatic updates to 2.0.0.4, Thunderbird says there is a message in Concept. I do know that isn't the case. The only used folders are Inbox and Outbox.

No concepts over here...? Or is that a (mis-)translation from something? In what way does it "say" there is a message there?
I use the Dutch version of Thunderbird on Mac OS 10.4.10 Under "Postvak IN" (Inbox) stands "Concepten" (concepts). The text is literly >Concepten (1)<. It is in bold. There is absolutely no message in concepts, altough Thunderbird says there is.

What is your "Auto Save" setting? (in Tools/Options/Composition/General)
Upgraded to Tb 2.0.0.4 from which version of Thunderbird?
You can always try going to properties of the Drafts folder, and do "Rebuild index". If that doesn't help, delete the Drafts.msf file (found in your profile folder).
To WADA: Thunderbird > Preferences > Compose > General > Auto Save is disabled. The update goes automatically. 2.2 MB bugfix (Macintosh Universal Binary NL) from 2.0 to 2.0.0.4

I deleted Drafts in User > Library > Thunderbird > Profiles > Directoryname > Mail > POP inbox > Drafts. Results: After a test to put a message in Draft, Thunderbird created a Directory concepten. Yes in Dutch. In User > Library > Thunderbird > Profiles > Directoryname > Mail > POP inbox came also a directory concepten instead of Drafts.

My answer to the problem was to copie Thunderbird from the server and copie the neccesay files as backup. Then delete Thunderbird completely and reinstall the new copie through Thunderbird 1.5.0.10 I tried first with reinstall 2.0 but the draftsfolder wasn't there  nor in User > Library > Thunderbird > Profiles > Directoryname > Mail > POP inbox, so then I reinstalled with 1.5.0.10 Now everything is like it should be.

"directory concepten instead of Drafts"? I assume "file concepten".

Tb 2.0.0.0 or before looks to use "Drafts" as file name for localized "concepten" folder, but Tb 2.0.0.4 looks to use localized "concepten" as file name for localized "concepten" folder.
Similar phenomenon was observed in some old Japanese versions of TB, and it caused some problems, and Japanese version of Tb currently doesn't change file name for standard mail folders, then Tb uses "Drafts" as file name insead of localized Japanese word.
Over localization in Dutch version of Tb 2.0.0.4?

Is filename for Drafts "concepten"? Or "Concepten"?
Do you use extention(add-on) or non-standard Theme?
Does filename of "concepten" occur on Tb 2.0.0.4 even when new profile created by Tb 2.0.0.4?
Following is localized mail folder name(not folder on Finder) of Tb for nl.
What file name is used currently for other mail folder than Drafts folder?
When was the file created?

Just received a mail from Joshua's post. I completely forgot this bugreport. Now I use the latest bugfix without any problems.
