in bug 478422 the user "removed Lightning version 0.9 add-on and then reinstalled. This seems to be a work-around."
does that work for you?
if not, does it work if you go back to lightning .8?
Severity: normal → critical
Summary: Lightning won't load → Lightning won't load after updating to 0.9 "An error was encountered preparing the calendar located at moz-profile-calendar:// for use. It will not be available"
Version: unspecified → Lightning 0.9

I've quite the same bug using Lightning 0.9 with Thunderbird Portable (2.0.0.23) on an USB key (windows XP).
When launching, I got an error message:
"An error was encountered preparing the calendar located at moz-abdirectory:// for use. It will not be available."
Detals:
Error number: 0xTypeError: Component.classes["@mozilla.org/calendar/calendar;1?type="+type] has no properties
Description: TypeError: Component.classes["@mozilla.org/calendar/calendar;1?type="+type] has no properties
And in console, I've (H:\ is my key drive):
Error: Assert failed: An error was encountered preparing the calendar located at moz-abdirectory:// for use. It will not be available.

Seems that you created a calendar using the ThunderBirthDay extension and disabled/removed the extension without removing the calendar in Lightning first. Reinstall ThunderBirthDay, remove the calendar from Lightning, uninstall ThunderBirthDay.
