An empty line is missing in the middle, as there used to be in TB2, to avoid signature and quoted message to be stuck too much. Reproducible: Always
Assuming the "reply above the quote" and "signature below my reply" scenario, are you talking about the space between the cursor and the start of the signature or the space between signature and quote? The latter was reduced on purpose by bug 428040.

Actually, I have to double check, as it may be linked to an extension that updated the reply configuration. I have to test on a new profile at work (I just had tested in safe-mode but on a migrated profile) because here at home I do not have the issue. More news tomorrow.
Sorry, you can cancel this bug: it was because Signature was to be set below my reply and reply above the quote. Moving signature below the quote has solved my problem. Thanks
