There's currently no possibility to set a reminder for "when the event starts" or alternatively for "0 Min. before the event starts".
(The mimimun for a reminder is currently only "1 Min. before the event starts".)

Maybe you are using an old nightly build of Lightning? This feature has been added with Bug 490169 several weeks ago and I see the corresponding menu option in Lightning 1.0b2pre (BuildID: 20100329044457).
You are right; works with the latest Nightly. Sorry.
