Using the Akismet API was brought up as a suggestion in bug 724882. This appears to be the best solution to our problem, since it is a proven system to fight spam before it's posted. The only downside is that it costs money. I don't think we get that many new comments per month, so maybe it'll be relatively cheap. Here are the pricing options: https://akismet.com/signup/

I talked this over with fligtar and it doesn't look like we'll do anything with Akismet in the future. I also think that whatever we end up doing for comment spam should be focused on the Marketplace and not AMO.
