The Back and Forward buttons move back and forward in your history of recently viewed messages. When you just start Thunderbird, the list is empty, so the buttons are greyed out. Maybe you want the Previous and Next buttons instead? They go to the previous and next message from the thread pane...
Can you check the menu under Go | Forward and Back. Are those options greyed out too?

wfm on WinXP, TB 12.0.1 with correct behaviour as described in comment 1. next and previous buttons, too. next goes to next unread msg, previous to previous unread, if there is none such, to first msg in msg list. If that's a problem (don't think so), would need new bug. Can someone double-check on Linux?
Summary: the radio buttons ¨BACK' and "FORWARD" are gray-out → toolbar buttons "BACK" and "FORWARD" are disabled (greyed-out)
