Soundcloud uses Flash to play audio, but when plugins.click_to_play is enabled, the area normally occupied by the flash widget is suspiciously empty.
Jaws is on vacation, so I'm setting needinfo for when he gets back.

This looks more like a usage/evangelism issue on that specific site. 
The actual Soundcloud sites handle Flash blockers graciously, see e.g.:
https://soundcloud.com/djshadow

The actual Flash instance is just in a 8x8 element at the left bottom of the page (#sm2-container / #sm2movie) and area that is staying blank is just non-Flash image & control elements.
So the actual player elements are defined in a 
> <script id='playerTemplate' type='text/x-jquery-tmpl'>
... which doesn't get inserted if Flash is blocked.
I'm not sure if the actual site or the soundcloud API are supposed to handle those templates.
