The setup:
Alice is running Windows 10 using Firefox Nightly
Bob is running using Firefox Nightly.

Bob and Alice enter a WebRTC call. Alice can only hear Bob when she is self muted (strange), and Bob can only hear Alice when she is unmuted (normal).

When Alice switches to release, everything works as expected.

I can reproduce this using https://appear.in and https://riot.im. I will post a regression range.

Mozregression narrowed it to Bug 1531833, which appears to be a Cubeb update, a change to use the default communications device on Windows, and more.

My default communications device was set to my Dell monitor, which has no attached speakers. Changing the default communications device to my normal speakers fixed the self mute problem. I am not sure if that is really the correct behavior though. Paul, would you mind taking a look?
