The Bugbug bot thinks this bug should belong to the 'Toolkit::Picture-in-Picture' component, and is moving the bug to that component. Please revert this change in case you think the bot is wrong.

Are you referring to the PiP button not being clickable on some Twitch streams? Or does the PiP button not show up at all?
I suspect this is probably due to Twitch sometimes injecting extensions overlays on top of the original video element. The PiP button expects to detect mouse events over the video elements and we use this to determine when to show it. I imagine these extension overlays interfere with this detection.
A workaround for this is to use the keyboard shortcut or opening the PiP window. On Windows, this is Ctrl + Shift + ].
I believe this is related to a fix we'd need to make in Bug 1539652, which requires some help from the DOM folks. Cc'ing Ania here just in case she wants to take a look and re-prioritize this.
