Starting any download (clicking on a link, and then clicking either Save or Open in the dialog) does not make it appear in the download manager, nor does it download the file.
Reproducible: Always
Steps to Reproduce:
1. Click on a link to a file
2. Select either Download or Open
3. Open the download manager
Actual Results:
The file is not added to the download manager
Expected Results:
The file is added to the download manager and downloaded
I also got https://bugzilla.mozilla.org/show_bug.cgi?id=371602 at the same time after updating. They may be linked.

Most probably downloads.rdf was corrupted at the same time as localstore.rdf (bug 371602). You should try renaming downloads.rdf from your profile (http://kb.mozillazine.org/Profile) while Firefox is closed.
If that fixes you up then this bug is a duplicate of bug 358914 - please attach the original downloads.rdf there (provided it doesn't contain anything private).

A malformed rdf file shouldn't cause it to not download still.
