Steps to reproduce:
youtube videos don't play. Screen is black. First noticed this with Version 15. I can still download the videos with the add-on Download Helper. Disabling Download Helper does not solve problem of videos not playing.

With Firefox closed try deleting pluginreg.dat file your Profile.

Did you close firefox before deleting the file ?
Have you tried the Firefox safemode ?

On testing it works in both current Chrome and Opera. I had Firefox 10 as well, where it *doesn't* work.

On Youtube are you 'opted in' on the HTML5 test? Opting out vids play fine with standard Flash, but not in HTML5.
I get this error in the console:

Ok so, weird but true:
1) I wasn't in the HTML5 trial and it didn't work
2) I joined the trial and it didn't work
3) I *left* the trial and it did work
So there's something messed up with HTML5 video on YouTube somehow.

For what it's worth, someone else confirmed that the above "join trial/leave trial" thing fixes it. No clue what that all means, but there you have it.

Videos don't display even in safe mode, never opted in for HTML 5,earlier versions of Minefield displayed videos fine, problem has only been the last couple of weeks.

I just tried http://www.youtube.com/watch?v=Bx5Sc3vWefE on WinXP SP3 on Firefox 12.0 release version, and its black, no video playback.
The even more looks like a YouTube issue to me...
This system won't support HTML5 due to the el-cheapo Intel Video Chipset, so its never been enabled, and won't enable.

The joining and leaving of the Youtube html5 beta sets a cookie.
It looks like a youtube issue if that makes a difference.
Deb: Does it stop working again if you delete the youtube cookies in Firefox ?
(Tools/options/privacy/dropdown:use custom settings-> Show cookies ...)

You could also try to override the current UA with the old format.

You system would not use hardware acceleration or webgl but html5 videos (webM or Og Theora) should work without problems.

The video that Deb mentioned and we have been using to test with, is failing across several releases at least, 10, my machine at work is on 12, and Nightly builds.
The recent UA change is only in Nightly builds so I don't think we can blame UA sniffing.
I also tested this morning with UA override pref and set it to 12.0 release UA and no change, video did not play, on my home system, Win7 x64 latest Nightly m-c build.

I couldn't reproduce this with Seamonkey trunk because I already had the youtube cookies set.
I can confirm that the issue with Firefox12 that I have for testing with a new profile.
Enabling html5 beta and disabling fixes this and just removing the cookies breaks it again !

I tried adding Alice0775's user_pref setting to user.js and it has no effect on my machine. I'm running this morning's build on Nightly. I even tried adding a general.useragent.override string to about:config without any effect. If a fix was pushed to Nightly it hasn't shown up in this morning's build.

Push wasn't pushed to Nightly, it was fixed on Google/Youtube's end. Their fix works for me here, and once HTML5 are working as well as the test video we were using to test with.
You may need to just delete your Youtube cookies or Google, or 'both'.. and try again, o the change has not propagated through all the servers yet.

Try deleting your YT cookies and try again. You could also try toggling the opt-in/opt-out of HTML5 test.

GOOD NEWS! an upgrade to Adobe Flash has solved this problem for me. You Tube and other flash video sites are all working now.
