Steps to reproduce:
Compose a message with addresses for Cc and Reply-To without using the mouse.
Actual results:
Switching from Thunderbird 68.x to 78.3.1, in the composer window, CC and Reply-To fields are not present by default, and buttons have to be clicked with the mouse to enable them.
I couldn’t find a configuration option to enable fields by default.
Expected results:
The user should be able to configure the message composer to have certain fields enabled by default.

Not true :-) Shift+Tab a few times will do the trick, or you could select recipients and press context menu key, then "move to cc/bcc".
Also note bug 1667692 which explores having dedicated keyboard shortcuts.

:
To add one or more recipients of another type, use the Cc, Bcc, or >> buttons on top of the To: addressing field. You can also hold down the Shift key and press Tab as often as required to navigate to these buttons.

Good point! I'm working on that in bug 1616514 (option to show CC/BCC by default).
As for Reply-to, you could enable auto-Reply-To for your identity in account settings.
