User Agent: Mozilla/5.0 (Windows NT 10.0; Win64; x64; rv:94.0) Gecko/20100101 Firefox/94.0
Steps to reproduce:
Go to Edit:Find:Search Messages
Change from "Search for messages in: (Inbox on my account)" to "Sent"
Change from "Subject" to ... Cannot choose "Body" because it is no longer available.
Actual results:
Nothing, not available.
Expected results:
Search on "Body" should be available.
WORKAROUND:
You can trick Thunderbird by searching your full account, and choose "Body", then switch search folders after. The "body search" stays.

Thunderbird 91.2.0 from Fedora on Fedora 34 Workstation.
Selected the default POP3 accounts Inbox.
Selected Edit > Find > Search Messages.
Switched to the Sent folder.
Changed from "Subject" and I see "Body" as a selection.
Switched to one of my IMAP accounts Inbox.
Selected Edit > Find > Search Messages.
The "Body" item is not available.
Probably because none of that account's folders are synchronized to store messages on my computer.
Selected "Run search on server" and it is now available for all folders.

I never even noticed that checkbox. Why would anyone ever uncheck that? Wouldn't it be easier to just run it the way it seems intuitively?

I agree with Walt, there must be a good reason why you are not seeing body. My guess is Bug 543416 - advanced search messages option to search in message body is not available for local search (run server-side searches w/o IMAP synchronization not obvious, won't stick)

Searching "online" means searching the server, which is slower. The default is to search locally, which is why the box is not checked
Wayne, what do you mean with
run server-side searches w/o IMAP synchronization not obvious, won't stick
Take a look at bug 564168 comment 4 (made 10 years ago).
