Took a quick peek at the DOM, and it seems that the video is loaded within an iframe. As observed in Bug 1757772, we can't launch PiP for videos in an iframe for the time being. Interestingly enough though, the pip toggle still appears in the case of twitchrivals when hovering the mouse cursor from the bottom.
Let me look into the case for supporting websites that load videos within an iframe, since this Twitch Rival problem is upsetting. Bug 1757772 has a priority of P5, but perhaps it should be bumped up as a result of the investigation.

I believe this is a duplicate of bug 1655802. They're both about extensions on Twitch videos that block the user from clicking the in-video PiP button.
