I confirmed on 5.0b2pre, 20110602000050.
The toolbar and its items have transparency on Windows 7. But it's hard to distinguish the buttons if back of the window is black.
I think that basically, toolbar shouldn't be on the glass. Otherwise, the buttons shouldn't have transparency.

Confirming. Here are some screenshots; you can see that if you've got a dark background and/or desktop icons in the area where the menu and toolbar are, both become quite difficult to read. A dark background is not the default, but Windows packs icons to the upper left, so that part, at least, will be a common issue. Even more irritating is when there is a rapidly-updating window behind the Thunderbird window.
My exposure to Windows 7 is pretty limited, but it seems to me that most Aero-aware applications do not make toolbars or menu bars partially transparent.

Retitling (this affects the menu bar too). This appears to be a regression from bugs 588007 and/or 545557.
