Steps to reproduce:
Wrote an email with "autosave mode" on.
Actual results:
My email got saved in the Draft folder once. Then again, as a duplicate! After sending the email, only one draft email is erased, the other duplicated clutter the Draft folder and one must remove them by hand.
Expected results:
Only one draft email should have been saved in the Draft folder. Duplicates are apparently timestamped versions of the same email, kept as reference... Very strange way to backup work in progress.

Works for me. Draft is only saved once (I had set the autosave to 1 minute and checked):

Thanks for your help Hashem! I confirm that this draft backup problem does not show up every time your write an email (that's where lies the tricky part). My guess is that something in the email content causes this backup problem. (This is especially true, I think, when you forward some content that was not produced on the local TB client.) Maybe this is also a MS Windows-specific issue...

Thanks Ludovic! It's a local folder, on my PC hard drive. In fact, I'm not using IMAP at all, only POP. Thanks in advance for your help!

Do you have extension installed ? does it happens all the time or just some times ?

Yes Ludovic, I do have a few extensions installed: the default ones, some dictionaries (I use the French version of TB) and AdBlockerPlus. Nothing fancy, really (I can provide a list if you want).
The problem does not happen all the time. It seems to depend on the mail content (encoding?), moreover, I would say that's a recent bug (as far as I remember, it appeared with Thunderbird v4). Thanks in advance!

And of course I forgot to ask if you had an anti viral solution running on your machine, if that's the case is quarantine enabled in Thunderbird ?

Thanks Ludovic! Yes, I am using F-Secure Internet Security 2012 with email protection activated. But I don't know how to check for this quarantine feature... Could this be the problem? Thanks in advance!

Tools -> options -> security -> antivirus , is that checked ?

Possibly following.
(1) draft-v1 is saved at offset=X of file named Drafts.
(2) auto-compact runs silently. draft-v1 is shifted to offsert=Y(Y<X).
(3) draft-v2 is needed, then Tb tries to delete draft-v1 at offset=X.
because draft-v1 is already moved, delete of drat-v1 is impossible.
(4) draft-v2 is saved at offset=Z
=> draft-v1 at offset=Y, draft-v2 at offset=Z
(5) Send => draft-v2 at offset=Z is deleted.
This is known phenomenon/issue.
If local mail folder, "Order Received" column is "offset in file". Show "Order Received" column and check column value of relevant draft mails.
"Dialog before start of auto-compact" is shown if mail.purge.ask=true is set(restart of Tb is mandatory after setting change). Do you enable auto-compact? If yes, is "Dialog before start of auto-compact" shown before your problem occurs?
Anyway, please rule out known phenomenon/issue due to auto-compact first.

Dear Wada, thanks a lot for your message. Yes, I do use auto-compact to save space, but no dialog shows up before auto-compact starts, it's done in the background before/after downloading new mail (I don't remember exactly when). This could indeed be the source of my problem. I will switch it off for a while and see how it goes... I keep you informed. Thanks again!

Dear Wada, as promised, here is some feedback. After several days without auto-compact, the problem I reported has disappeared completely. I guess your explanation is right: this bug is a well-known issue... Should we close my report and/or merge it with another report? Please don't hesitate to ask me for more details.
