Steps to reproduce:
1. Start the Thunderburd. Version 24, update 24.0.1.
2. CTRL + N - new message
3. Start to write a message.
4. Make a typo. The misspelled word is underlined now.
5. IGNORE IT (DO NOT CORRECT) and continue to write without typos some a few words.
6. Make a typo again and ERASE THIS WORD OR THE MISSPELLED PART of this word with the BACKSPACE key.
Actual results:
The first misspelled (and NOT CORRECTED BEFORE) word (or words) is now NOT UNDERLINED anymore.
Expected results:
The misspelled words should stay underlined until ending the messages independently on the fact whether the misspelled words are corrected immediately after they occur or the errors are ignored and are corrected after ending the hole writing process. So they should be underlined during the hole cycle of writing, from beginning to the end.

I am not able to reproduce your problem here. If you could supply a testcase eml that would be quite helpful. (It could be dependent on font selection and sizing within a specific email)

Unfortunately, I do not see anything unusual in that mail, nor can I reproduce your problem.
Tested with current trunk as well as 24.0.1

STR from email:
Dear Mr. Bell,
in respons to yur request we are glad to sey that yur project was accepted. Concerning this we would discuss som points we see important for our futher project and,
Steps to reproduce.
1. Spell checking dictionary is English.
2. Start to write the text above. Write normally and Make EXACTLY THE SAME errors in the words above, they are marked as red. DO NOT correct the misspelled words during the writing process.
3. You see the misspelled word as underlined.
4. Now completely erase only the word futher till the word our with BACKSPACE KEY.
5. Type the word futher with an error again. Erase it again. Repeat this step (step 4) 2-4 times.
6. All the red marked words are now NOT UNDERLINED anymore, at least in my case. If the word futher is still misspelled you can see only this misspelled and underlined word, but NOT the others written before.
Unfortunately, following your steps, I still did not see your problem.
It may be something specific to your setup or profile.
(for instance: Tools>>Options>>Composition>>General The time period for autosave as draft)
