You're supposed to be able to right-click a separator in the bookmarks toolbar and delete it. It's very difficult to get this context menu to come up; you need to click perfectly on a specific pixel it seems. The clickable area for bringing up the context menu needs to be a lot bigger for a separator.
Component: Toolbars and Customization → Bookmarks & History
Which OS? On Windows I don't have big trouble clicking on it, even if it's really not that large. It may be useful to increase padding on separators.

I managed to reproduce the issue using an older version of Nightly on macOS 10.13. I retested everything on the same OS using Firefox 84.0 and Nightly 85.0a1. The issue is not reproducing anymore.
However, this issue is not fixed for Mac OS 10.11 (or older versions) because I can still reproduce it on ESR 78.6.0 (older versions of Firefox don't work anymore on those os's). Maybe you can consider making a patch for esr builds, too.
